Composition and method for high efficiency chromatography

Abstract

The invention provides novel compositions, columns and methods for improved chromatographic separations. In particular, novel column packings are provided to improve overall chromatographic separation and efficiency. The invention provides a packing material having particles or mixtures of particles that are used for separation of small molecules, proteins or nucleic acids.

    A column for use in chromatographic separations, comprising:(a) a housing;and (b) a stationary phase disposed inside the housing, the stationary phase comprising a packing material comprising two or more components each having a particle size distribution and a mean particle diameter wherein the difference between any two mean diameters is less than or equal to 40% of the larger mean value.

    A column for use in chromatographic separations, comprising:(a) a housing;and (b) a stationary phase disposed inside the housing, the stationary phase comprising a first component having a particle number between 500 and 3000, and a particle mean diameter between 1 and 4 microns and a second component having a particle number between 500 and 3000, and a particle mean diameter between 1 and 4 microns.

    A composition of matter for use as a stationary phase in a chromatography packing material, said packing material comprising particles or mixtures of different porosity particles that have a bimodal number average particle size distribution in which the difference between the diameters at the two peaks of the distribution is less than or equal to 40% of the value of the average of the diameters at the two peaks.

    A method of chromatographic separation comprising:(a) contacting a sample to a stationary phase having a packing material comprising particles that have a particle size distribution in which the difference between the particle diameters at the two peaks of each distribution is less than or equal to 40% of the value of the average of the diameters of the two peaks;and (b) separating the sample components.

    A method for chromatographic separation comprising:(a) contacting a sample to a stationary phase, said stationary phase having a packing material comprising a first and second component, the number average particle size distribution of the first component being broader than the number average particle size distribution of the second component and wherein the maximum particle size diameter of the first component is approximate to the maximum particle size diameter of the second component.
