Methods for selectively stimulating proliferation of T cells

Abstract

Methods for inducing a population of T cells to proliferate are described which involve activating the T cells with a first agent which provides a primary activation signal to the T cells; and stimulating the activated T cells with a second agent which stimulates an accessory molecule on the surface of the T cells.

    An in vitro method for inducing a population of T cells to proliferate, comprising contacting a population of T cells with a surface having attached thereon:(a) a first agent which provides a primary activation signal to the T cells, thereby activating the T cells;and(b) a second agent which stimulates an accessory molecule on the surface of the T cells, thereby stimulating the activated T cells, the first and second agents thereby inducing the T cells to proliferate.

    The method of any one of claims 1 to 7, wherein the accessory molecule on the T cell is CD28.

    The method of daim 9, wherein the anti-CD3 antibody is one of OKT3 or G19-4, and the anti-CD28 antibody is one of 9.3 or EX5.3D10.

    The method of any one of claims 1 to 13, further comprising:monitoring proliferation of the T cells;and reactivating and restimulating the T cells with the first and second agents when the rate of T cell proliferation has decreased to induce further proliferation of the T cells.

    The method of any one of claims 1 to 16, wherein the population of T cells is obtainable from an individual infected with HIV.

    The method of any one of claims 1 to 16, wherein the T cells from an individual afflicted with an immunodeficiency associated with a genetic defect are genetically transduced to correct for the defect.

    The method of any one of claims 1 to 23, further comprising repeating the steps (a) - (b) to produce a population of T cells increased in number of from about 100- to about 100,000-fold of the original T cell population.
