VEGF receptor chimeras for the treatment of eye disorders characterized by vascular permeability

Abstract

Modified chimeric polypeptides with improved pharmacokinetics are disclosed useful for treating eye disorders, including age-related macular degeneration and diabetic retinopathy.

    Use of a fusion polypeptide capable of binding vascular endothelial growth factor (VEGF) in the manufacture of a medicament for treating an eye disorder or arthritis, wherein the fusion polypeptide consists essentially of an immunoglobulin-like (Ig) domain 2 of a first VEGF receptor and Ig domain 3 of a second VEGF receptor and a multimerizing component.

    Use according to any one of the preceding claims, wherein the fusion polypeptide is encoded by a nucleic acid sequence as set forth in (a) Figures 21A-21C, (b) Figures 22A-22C, (c) Figures 24A-24C or a nucleotide sequence which, as a result of the degeneracy of the genetic code, differs from the nucleotide sequence of (a), (b) or (c).

    Use according to any one of the preceding claims wherein the fusion polypeptide comprises the amino acid sequence set forth in Figures 21A-21 C, Figures 22A-22C or Figures 24A-24C.

    Use according to any one of the preceding claims, wherein the eye disorder is age related macular degeneration or diabetic retinopathy.

    Use according to any one of claims 1-7 wherein the arthritis is rheumatoid arthritis.

    Use of a fusion polypeptide capable of binding vascular endothelial growth factor (VEGF) in the manufacture of a medicament for use in treating an eye disorder, wherein the fusion polypeptide consists essentially of an immunoglobulin-like (Ig) domain 2 of a Flt1, an Ig domain 3 of a Flk1 or Flk4 and a multimerizing component, wherein the multimerizing component is an immunoglobulin domain.

    Use of a fusion polypeptide capable of binding vascular endothelial growth factor (VEGF) in the manufacture of a medicament for use in treating arthritis, wherein the fusion polypeptide consists essentially of an immunoglobulin-like (Ig) domain 2 of a Flt1, an Ig domain 3 of a Flk1 or Flk4 and a multimerizing component, wherein the multimerizing component is an immunoglobulin domain.
