    Claims of equivalent WO 03087390 A2 CLAIMS What is claimed is:1. A method for detecting an indicium of a disease in a heterogeneous biological sample, said method comprising the steps of: isolating a heterogeneous nucleic-acid sample comprising a CpG-containing target template from a heterogeneous biological sample;contacting said nucleic-acid sample with an agent that modifies unmethylated cytosine;performing an amplification reaction on said target template using at least one chimeric primer that comprises a first portion that is not specific for said target template and a second portion 3' to said first portion, said second portion being specific for said CpG-containing target template if said template comprises an unmodified cytosine at a predetermined position;and detecting the presence of an amplification product as an indicium of a disease.

    A method for detecting an indicium of colorectal cancer in stool, said method comprising the steps of:isolating a heterogeneous nucleic-acid sample comprising a CpG-containing target template from stool;contacting said nucleic-acid sample with an agent that modifies unmethylated cytosine;performing an amplification reaction on said target template using at least one chimeric primer that comprises a first portion that is not specific for said target template and a second portion 3 ' to said first portion, said second portion being specific for said CpG-containing target template if said template comprises an unmodified cytosine at a predetermined position;and detecting the presence of an amplification product as an indicium of colorectal cancer.

    A method of screening a population of patients for colorectal cancer, the method comprising the steps of:isolating a plurality of target nucleic acid templates from each patient;and assaying each of said templates for the presence of hypermethylation according to the method of claim 2, wherein a patient is diagnosed as having an indicium of colorectal cancer if at least one of said templates from said patient is hypermethylated.

    A method for detecting an indicium of a disease in a heterogeneous biological sample, said method comprising the steps of:isolating a heterogeneous nucleic-acid sample comprising a CpG-containing target template from a heterogeneous biological sample;contacting said nucleic-acid sample with an agent that modifies unmethylated cytosine;performing an amplification reaction on said target template using at least one chimeric primer that comprises a first portion that is not specific for said target template and a second portion 3' to said first portion, said second portion being specific for said CpG-containing target template if said template comprises a modified cytosine at a predetermined position;and detecting the presence of an amplification product as an indicium of a disease.
