Performing polymerase chain reaction

Abstract

Disposable polypropylene tubes are used as reaction containers for performing polymerase chain reaction (PCR) on samples of nucleic acids. The tubes have a lower conical portion and an upper cylindrical portion. The tubes are capped in use. The lower portion contacts a correspondingly shaped hole in a heating block and has a substantially thinner wall than that of the upper portion of the tube. Downward pressure on the capped tube ensures a tight fit of tube in the heating block and ensures rapid heat exchange between the block and sample in the tube.

    A method for performing a polymerase chain reaction (PCR) comprising providing a thermocycler apparatus comprising a sample block containing one or more sample wells, one or more capped reaction tubes disposed in the one or more sample wells, and a heated platen for contacting the caps of the one or more reaction tubes, wherein each tube comprises a PCR reaction mixture,subjecting the reaction mixture(s) to a PCR protocol comprising a plurality of repeated cycles by cycling the temperature of the sample block, each cycle comprising at least a primer extension temperature and a denaturation temperature, such that the heated platen is kept at a temperature of from 94°C to 110°C and remains in contact with the caps of the one or more tubes during the PCR protocol, whereby condensation of water on the undersides of the caps is prevented.

    The method of any one of the preceding claims, wherein the PCR protocol comprises 25 to 30 repeated cycles.

    The method of any one of the preceding claims, wherein the platen provides a downward force on the caps such that the tubes are pressed firmly into their respective sample wells.

    The method of any one of the preceding claims, wherein the downward force is at least 30 grams.

    The method of any one of the preceding claims, wherein the downward force is at least 50 grams.

    The method of any one of the preceding claims, wherein the downward force is between 50 and 100 grams.

    A thermocycler apparatus suitable for performing the polymerase chain reaction comprising a metal sample block having an array of spaced-apart sample wells each of which has an inside surface, said wells being provided with one or more capped sample tubes each containing a sample mixture placed in a microtiter plate having an uppermost edge, which plate is seated on said sample block, anda cover to enclose said capped sample tubes, which cover comprises a flat, horizontal rectangular portion and downwardly projecting skirt portions along the periphery thereof and further comprises a device for heating at least the underside of said horizontal portion, said cover being dimensioned to contact said sample block and to enclose said microtiter plate and capped sample tubes on said sample block when the tops of the caps on said sample tubes deform, due to the application of heat and a downwardly directed force on said cover.
