Abstract

A method and coating for reducing the release rate of an active agent from an implantable device, such as a stent, is disclosed.

    A stent comprising a radially expandable body and a coating covering at least a portion of the body, the coating comprising a reservoir layer comprising:(a) a polymer, and (b) 40-O-(2-hydroxy)ethyl-rapamycin, 40-O-(3-hydroxy)propyl-rapamycin or 40-O-[2-(2-hydroxy)ethoxy]ethyl-rapamycin;wherein the release rate of the 40-O-(2-hydroxy)ethyl-rapamycin, 40-O-(3-hydroxy)propyl-rapamycin or 40-O-[2-(2-hydroxy)ethoxy]ethyl-rapamycin in 24 hours after the implantation of the stent is less than about 50% of the total amount present in the coating.

    Use of 40-O-(2-hydroxy)ethyl-rapamycin, 40-O-(3-hydroxy)propyl-rapamycin or 40-O-[2-(2-hydroxy)ethoxy]ethyl-rapamycin in the manufacture of a stent for inhibiting or eliminating the development of restenosis following a stent placement procedure, wherein the drug is contained in a polymer coating on the stent, and wherein the stent is capable of eluting 40-O-(2-hydroxy)ethyl-rapamycin, 40-O-(3-hydroxy)propyl-rapamycin or 40-O-[2-(2-hydroxy)ethoxy]ethyl-rapamycin at a release rate of less than 50% of the total amount of the drug carried by the stent in a 24 hour period following implantation.

    A process for producing a stent which is capable of drug delivery, the process comprising coating a stent with a polymer comprising 40-O-(2-hydroxy)ethyl-rapamycin, 40-O-(3-hydroxy)propyl-rapamycin or 40-O-[2-(2-hydroxy)ethoxy]ethyl-rapamycin wherein the coating has an in vivo release rate of less than 50% of the total amount of the 40-O-(2-hydroxy)ethyl-rapamycin, 40-O-(3-hydroxy)propyl-rapamycin or 40-O-[2-(2-hydroxy)ethoxy]ethyl-rapamycin, in a 24 hour period.
