17 claims: 11 independent, 6 dependent

  1. 1
An immunoglobulin-binding protein capable of binding to regions of an immunoglobulin molecule other than the complementarity determining regions (CDR), wherein said protein comprises two or more repetitive units as defined by SEQ ID NO. 1 or 2, in which the amino acid residue at position 23 in each unit is a threonine.
  2. 3
A protein according to anyone of the preceding claims, wherein the units are linked by elements comprised of up to about 15 amino acids.
  3. 4
A protein according to any one of the preceding claims, which also comprises one or more repetitive units comprised of the E, D, A, B, and C domains of Staphylococcus protein A.
  4. 5
A protein according to any one of the preceding claims, which is a tetramer.
  5. 7
A nucleic acid encoding a protein as defined in any one of claims 1-6.
  6. 9
A matrix for affinity chromatography, wherein a plurality of ligands comprising immunoglobulin-binding proteins according to any one of claims 1-6 have been coupled to a solid support.
  7. 11
A matrix according to 9 or 10, wherein the ligands have been coupled to the support by thioether bonding.
  8. 12
A matrix according to any one of claims 9-11, wherein the support is a polysaccharide material.
  9. 13
A matrix according to any one of claims 9-12, which provides selective binding of an immunoglobulin selected from the group that consists of IgG, IgA and IgM, preferably IgG.
  10. 14
Use of a matrix according to any one of claims 9-13 in chromatographic isolation of an immunoglobulin, which use comprises the steps of adsorbing immunoglobulin to the matrix;eluting said immunoglobulin;cleaning the matrix with alkali;adsorbing a immunoglobulin to the cleaned matrix;and eluting the immunoglobulin.
  11. 16
A process of chromatography, wherein at least one target compound is separated from a liquid by adsorption to a protein according to any one of claims 1-6, or to a matrix according to anyone of claims 9-13.
