8 claims: 2 independent, 6 dependent

  1. 1
A method of analyzing an analyte in a urine test sample on a matrix of an absorbant material through which the urine test sample, analyte and labeled antibody can flow by capillarity, characterized by the steps of:a) combining a labeled antibody specific to the analyte with the urine;b) determining the concentration of the analyte in the urine by measuring the magnitude of the response from the detectable label;and c) maintaining the concentration of urea in the sample sufficiently high so that it does not affect the binding of the analyte and antibodies, and therefore the accuracy of the assay.
  2. 2
The method of Claim 1 wherein the analyte is doexypyridinoline and sufficient urine is combined with the urine test sample to provide a urea concentration therein of at least 225 mM.
  3. 3
The method of Claims 1 and 2 wherein the matrix of absorbant material comprises nitrocellulose, a polysulfone a polycarboxylic acid or filter paper.
  4. 4
The method of Claims 1 to 3 wherein the matrix is in the form of a strip which strip has a buffer containing pad upstream from the labeled antibodies and the urea necessary to increase the concentration of urea in the urine test sample is stored in this buffer pad.
  5. 5
The method of Claims 1 to 4 wherein the results of the assay are obtained by comparing the magnitude of the response from the detectable with responses obtaining from calibrators containing similar amounts of additional urea.
  6. 6
A test strip for the determination of an analyte in a urine test sample which comprises a matrix of an absorbant material through which the urine, analyte and labeled antibodies to the analyte can flow by capillarity which strip has a region containing mobile, labeled antibodies specific for analyte, and a region containing dry urea in sufficient quantity so that when the urea is rehydrated upon contact its concentration does not affect the binding between the analyte and the labeled antibodies, wherein the region of the strip containing the urea is located in an area of the strip which is upstream from the region which contains the mobile, labeled antibodies.
  7. 7
The strip of Claim 6 which contains a buffer containing pad and the urea is stored in this buffer pad.
  8. 8
The test strip of Claim 7 wherein the analyte is deoxypyridinoline and the the concentration of urea in the urine test sample is greater than 225mM.
