Bioabsorbable branched polymers containing units derived from dioxanone and medical/surgical devices manufactured therefrom

Abstract

Star polymers of soft segment forming monomers are useful in forming surgical devices. The star polymers can be endcapped with isocyanate, mixed with a filler and/or cross-linked. The polymer compositions are useful, for example, as fiber coatings, surgical adhesives or bone putty, or tissue growth substrate.

    A biocompatible composition comprising:a) a star polymer of the general formula:         CH 2 OR 1 -(CHOR 2 )-(CHOR 3 )-(CHOR 4 )...(CHOR n )-CH 2 OR n+1 wherein: n equals 1 to 13;R 1 , R 2 ...R n+1 are the same or different and selected from the group of a hydrogen atom or (Z) m wherein Z comprises repeating units including one or more member selected from the group consisting of: wherein p is 3 to 8 and each R' may be the same or different and are individually selected from the group consisting of hydrogen and alkyl having from 1 to 5 carbon atoms, such that at least three of said R 1 , R 2 ...R n+1 groups are other than hydrogen;m is sufficient such that the star polymer has an inherent viscosity in HFPI at 25°C between about 0.01 and about 0.5 dl/gm;and the m's for each (Z) group may be the same or different, at least one of said (Z)m groups being encapped with an isocyanate;and b) a polysaccharide modified to contain a charge inducing substituent.

    A composition an in claim 1 wherein the polymer is at least partially cross-linked through the isocyanate.

    An adhesive for hard tissue comprising a fluid composition containing an isocyanate endcapped absorbable star polymer, the polymer being capable of undergoing cross-linking when exposed to water thereby curing to provide a solid material.

    A surgical device comprising:a) a fusion implant;and b) a bone-growth-inducing substance applied to the fusion implant, the bone-growth-inducing substance comprising a composition containing a synthetic bioabsorbable polymer and a filler, wherein the weight ratio of polymer to filler is in the range of about 35:65 to about 15:18.

    A bone substitute material comprising:an isocyanate endcapped bioabsorbable star polymer;a filler;and a polysaccharide modified to contain a charge inducing substituent;wherein the bone substitute material is packaged in a moisture impervious package in a substantially dry state whereby crosslinking of the encapped bioabsorbable star polymer substantially inhibited prior to use of the bone substitute material.
