Microporous polysulfone supports suitable for removal of low density lipoprotein-cholesterol.

Abstract

The invention relates to a microporous plasmapheresis support having an immobilised affinity agent which is a polyanion bound directly and/or through an interaction with silica and/or calcium chloride to a microporous polysulfone structure and to a process for preparing such a support. The support is useful for the efficient removal of low density lipoprotein cholesterol complex (LDL-C) from whole blood.

    A support for binding low density lipoprotein cholesterol comprising a microporous polysulfone structure having an amount of a polyanion effective to bind low density lipoprotein cholesterol immobilised in a network interpenetrating the surface of said polysulfone structure.

    A support according to Claim 1 wherein said structure is a flat sheet membrane.

    A support according to any one of the preceding claims wherein the polyanion is polyacrylic acid or a polyalkacrylic acid containing from 1 to 4 carbon atoms in the alkyl group.

    A support for binding low density lipoprotein cholesterol consisting essentially of a microporous polysulfone structure having polyacrylic acid and a calcium ion-containing compound immobilised in a network interpenetrating the surface of said polysulfone structure, wherein the amount of polyacrylic acid is effective to bind low density lipoprotein cholesterol and the calcium ion-containing compound is in an amount effective to enhance the quantity of polyacrylic acid immobilised on the structure.

    A support for binding low density lipoprotein cholesterol consisting essentially of a microporous polysulfone structure having polyacrylic acid, a calcium ion-containing compound and silica immobilised in a network interpenetrating the surface of said polysulfone structure, wherein the amount of polyacrylic acid is effective to bind low density lipoprotein cholesterol and the calcium ion containing compound and silica are in an amount effective to enhance the quantity of polyacrylic acid immobilised on the structure.

    A process for preparing a support which is capable of binding low density lipoprotein cholesterol, which process comprises (a) casting the support having a microporous polysulfone structure from a casting solution comprising from 8 to 22 wt. % of a polysulfone polymer and optionally up to 10 wt. % silica;(b) submerging said support in an acidic solution comprising polyacrylic acid and optionally up to 3 wt. % calcium chloride;(c) immobilising polyacrylic acid to said support by heating under increased pressure the support submerged in step (b);and (d) annealing the support of step (c) by heating under increased pressure in water.

    A process according to any one of claims 10 to 13 wherein the membrane is vacuum degassed between step (c) and step (d).

    A process according to any one of claims 10 to 14 in which steps (c) and (d) are carried out under increased pressure at from 140 to 280 kPa (20 to 40 psi).

    A process according to any one of claims 10 to 16 wherein the membrane is soaked in a basic solution between step (a) and step (b).

    A process according to any one of claims 10 to 17 which further comprises:(e) adjusting the solution of step (d) to a pH in the range of 7.5 to 10.5;and (f) drying the membrane in the presence of a salt and a surfactant.

    Apparatus for the separation of low density lipoprotein from blood which comprises a support according to any one of claims 1 to 9.
