Amine rich fluorinated polyurethaneureas and their use in a method to immobilize an antithrombogenic agent on a device surface.

Abstract

A thermoplastic polyurethaneurea having free amino groups and comprising a product from the reaction of a diisocyanate, a polyamine and a misture of polyols comprising a fluorinated polyol and a non-fluorinated polyol selected from polyalkyleneoxide polyols and polyester polyols. A medical device having a hemocompatible surface comprising a shaped polymeric article having thereon a coating of a thermoplastic polyurethaneurea as claimed in Claim 1, and an antithrombogenic agent covalently affixed to amino groups of the polyurethaneurea coating.

    A method for preparing a medical device having a hemocompatible surface comprising:a) reacting a diisocyanate with a mixture of polyols comprising a fluorinated polyol and a non-fluorinated polyol selected from polyalkyleneoxide polyols and polyester polyols to give a prepolymer having terminal isocyanate groups;b) coating a polymeric support structure with the prepolymer to give a support sructure having isocyanate groups on its surface;c) reacting said support structure having isocyanate groups on its surface with a polyamine to give a support structure having free amino groups on its surface;andd) reacting said free amino groups with an activated antithrombogenic agent whereby the antithrombogenic agent becomes covalently affixed to the support structure.

    A method for preparing a medical device having a hemocompatible surface comprising:a) reacting 4,4′-diphenylmethane diisocyanate with a mixture of polyols consisting of a fluorinated polyol and polytetramethylene ether glycol to give a prepolymer having terminal isocyanate groups;b) coating a polymeric support structure with the prepolymer to give a support structure having isocyanate groups on its surface;c) reacting the support structure having isocyanate groups on its surface with a polyamine to give a support structure having free amino groups on its surface;andd) reacting the free amino groups with aldehyde-activated heparin whereby the heparin becomes covalently affixed to the support structure.

    A method for preparing a medical device having a hemocompatible surface comprising:a) reacting a diisocyanate with a mixture of polyols comprising a fluorinated polyol and a non-fluorinated polyol selected from polyalkyleneoxide polyols and polyester polyols and a polyamine to give a fluorinated polyurethaneurea having free amino groups;b) coating a polymeric support structure with the polyurethaneurea to give a support structure having free amino groups on its surface;andc) reacting the free amino groups with an activated antithrombogenic agent whereby the antithrombogenic agent beoomes covalently affixed to the support structure.
