Gel filtration of heat treated factor VIII.

Abstract

Highly purified antihemophilic factor is produced by a process comprising a PEG precipitation step, a gel filtration step and a heat treating virus inactivation step. Al(OH)₃ adsorption and PEG precipitation carried out at room temperature allow processing to proceed directly to a gel filtration step.

    A process for the production of a concen­trate of antihemophilic factor (AHF) from cryoprecipitate comprising the steps of:(a) dissolving said cryo­precipitate;(b) removing non-AHF proteins by precipitation with polyethylene glycol (PEG);(c) heat treating said AHF for viral inactivation;then (d) passing said AHF through a gel filtration column containing a size exclusion resin to remove said chemical and to concentrate said AHF to at least 35 units of Factor VIII activity per ml of pooled concentrate.

    A process for the production of antihemo­philic factor concentrate (AHF) comprising the steps of:(a) preparing a cryoprecipitate;(b) removing non-AHF proteins by precipitation to form a solubilized AHF pool;(c) passing said pool through a gel filtration column;and (d) heating AHF obtained from said column in a solution in the presence of sucrose.
