Capillary tube with reduced protein interactions and controllable electroosmotic flow.

Abstract

A method for treating capillary walls exposable to protein solutes is provided that reduces interactions of the protein solutes with the wall and that permits control of electroosmotic flow within the capillaries by selection of solution pH. Thus, a capillary tube has an interfacial layer covalently bonded to the inner wall of the tube. This interfacial layer is effective to reduce interactions between the inner wall and protein solutes and preferably includes a hydratable amphoteric phase. The amphoteric phase is a reaction product of a protein, peptide or an ampholyte and an oxygen or nitrogen nucleophile. Electroosmotic flow magnitude and/or flow direction may be controlled when capillaries of the invention are used for electrophoretic separations.

    A capillary tube defining an inner wall, useful for electrophoretic separations of proteins in solution, comprising:an interfacial layer covalently bonded to the inner wall and effective to reduce interactions between the inner wall and protein solutes, the interfacial layer including a hydratable amphoteric phase.

    The capillary tube as in claim 1 wherein the amphoteric phase has a determinable isoelectric point and selectively permits electroosmotic flow control.

    The capillary tube as in claim 2 wherein the electroosmotic flow control is selected by solution pH.

    The capillary tube as in claim 3 wherein the amphoteric phase permits control of electroosmotic flow magnitude.

    The capillary tube as in claim 3 wherein the amphoteric phase permits control of electroosmotic flow direction.

    The capillary tube as in one of claims 1 to 5, wherein the amphoteric phase includes ionizable cationic and ionizable anionic species, the cationic species selected from the group consisting of amino, guanidinium, imidazolium and mixtures thereof, the anionic species being carboxyl groups.

    The capillary tube as in one of claims 1 to 5, wherein the amphoteric phase is a reaction product of a protein, peptide or an ampholyte and an oxygen or nitrogen nucleophile.

    A small bore capillary tube, useful for electrophoretic separations of protein solutes, comprising:a reduced interaction phase covalently bonded to the bore and effective to reduce interactions between the bore and protein solutes, the reduced interaction phase including an ionizable species having an acidic equilibrium or a basic equilibrium, the ionizable species permitting electroosmotic flow control by pH selection.

    The small bore capillary tube as in claim 8 wherein the ionizable species is an amino group and permits control of electroosmotic flow magnitude and direction.

    The small bore capillary tube as in claim 8 wherein the ionizable species is a carboxyl group and permits control of electroosmotic flow magnitude.
