Method for discriminating between intact and damaged cells in a sample.

Abstract

A method is disclosed for the discrimination between intact and damaged cells in a sample, and more particularly, the method uses a vital nucleic acid dye to selectively stain intact versus damaged cells which then may be counted and sorted by flow cytometry means. The method also may be used in conjunction with fluorescently labelled mononclonal antibodies to simultaneously identify cellular antigens.

    A method for discriminating between intact and damaged cells in a body fluid sample which comprises the steps of:1) taking a body fluid sample from an indi­vidual;2) adding a nucleic acid dye to said sample;and3) analyzing said sample in an automated instrument capable of passing the cells in a sample through a sensing region substantially one at a time and capable of detecting and recording both fluores­cence and light scattered in more than one direction by each cell passing through said sensing region.

    A method for discriminating between intact and damaged cells in a peripheral blood sample comprising the steps of:1) taking a sample of peripheral blood from an individual;2) adding at least one fluorescently labelled MAb to the sample wherein the fluorescent labels have peak emission spectra that are distinguishable from each other and from a nucleic acid dye;3) adding a fixative to the sample;4) adding a nucleic acid dye with a preference for DNA;and5) analyzing said sample in an automated instru­ment capable of passing the cells in the sample through a sensing region substantially one at a time and capable of detecting and recording both fluores­cence and light scattered in more than one direction by each cell passing through said sensing region.

    A method for discriminating between contact and damaged cells in a peripheral blood sample which comprises the steps of:1) taking a sample of peripheral blood from an individual;2) lysing the erythrocytes in said sample;3) adding at least one fluorescently labelled MAb to said sample, wherein the fluorescent labels have peak emission spectra different from each other and LDS-751;4) adding paraformaldehyde to the sample;5) adding LDS-751 to said sample;and6) analyzing said sample in a flow cytometer equipped with a single laser tuned to 488nm.

    A kit for discriminating between intact and damaged cells in a body fluid sample which comprises containers separately containing a nucleic acid dye and one or more MAbs.
