Improvements in the production of polypeptides.

Abstract

A novel process for the production of heterologous proteins including the use of certain transformed protease deficient yeast strains is provided. The invention concerns also said transformed yeast strains and methods for the production thereof.

Method for the production of a protein heterologous to yeast accord­ing to claim 12 wherein the hybrid vector consists of a yeast promoter operably linked to a first DNA sequence encoding a signal peptide linked in the proper reading frame to a second DNA sequence coding for said heterologous protein and a DNA sequence containing yeast transcription termination signals.

    A desulphatohirudin of the formula in which X₁ represents the dipeptide residue Val-Val, X₂, X₃ and X₄ are Lys, X₅ is His and X₆ is selected from the group consisting of Glu-Tyr-Lys-Arg, Ser-Phe-Arg-Tyr and Trp-Glu-Leu-Arg. 19. Method according to claim 16 for the preparation of a desulphato­hirudin of the formula in which X₁ represents the dipeptide residue Val-Val, X₂, X₃ and X₄ are Lys, X₅ is His and X₆ is selected from the group consisting of Glu-Tyr-Lys-Arg, Ser-Phe-Arg-Tyr and Trp-Glu-Leu-Arg.
