Apparatus and method for dilution and mixing of liquid samples.

Abstract

An apparatus for diluting a sample with a di­luent, comprising a fixed volume measuring chamber, a fixed volume receiving chamber and fluid receiving re­lationship to the measuring chamber, a gas vent in the receiving chamber, a stop flow junction between the measuring chamber and the receiving chamber, a sample application site in fluid donating relationship to the measuring chamber, wherein the vertical height differ­ence between the sample application site and the stop flow junction is insufficient to provide a hydrostatic pressure capable of overcoming backpressure at the stop flow junction when sample is applied to the sample ap­plication site, and the diluent application site and fluid donating relationship to the measuring chamber. Sample and diluent are added sequentially to the appar­atus of the invention to achieve dilution without requiring any intervening operations. In a second embodiment of the invention, a dilution and mixing cartridge comprising a sample application site, a mixing chamber in fluid receiving relationship to the sample application site, a diluent application site in fluid donating relationship to the mixing chamber, a mixture isolating chamber hydrostatically connected to the mixing chamber, and a first valve means selectively preventing flow from the diluent application site to the mixing chamber, and a mixture isolating chamber hydrostatically connected to the mixing chamber is provided along with a method for using this cartridge to sequentially dilute a sample with the same or different diluents.

    An apparatus for diluting a sample with a diluent comprising:a fixed volume measuring chamber;a fixed volume receiving chamber in fluid receiving relationship to said measuring chamber;a gas vent in said receiving chamber;a stop flow junction between said measuring chamber and said receiving chamber;a sample application site in fluid donating relationship to said measuring chamber, wherein the vertical height difference between said sample applica­tion site and said stop flow junction is insufficient to provide flow through said stop flow junction when sample is applied to said sample application site;and a diluent application site in fluid donating relationship to said measuring chamber.

    A method of diluting a sample with a diluent, comprising:adding a sample to a sample application site of an apparatus from which sample flows without the ap­plication of external energy, other than gravitational energy, into a fixed volume measuring chamber termin­ated by a stop flow junction which stops sample flow;adding a diluent to a diluent application site of said apparatus from which diluent is capable of flowing through said measuring chamber and stop flow junction and into a fixed volume measuring chamber;and starting flow at said stop flow junction, wherein diluent drives sample in said measuring chamber into said receiving chamber without the application of external energy other than gravitational energy.

    A dilution and mixing cartridge, comprising:a sample application site;a mixing chamber in fluid receiving relation­ship to said sample application site;a diluent application site in fluid donating relationship to said mixing chamber;a mixture isolating chamber hydrostatically connected to said mixing chamber;and first valve means selectively preventing flow between said mixing chamber and said mixture isolating chamber, whereby a measured representative sample of the contents of the mixing chamber can be selectively isolated and preserved in the mixture isolating chamber.

    A method for sequentially diluting a liquid sample with two liquid diluents, comprising:supplying a predetermined volume of said sam­ple and a predetermined volume of a first liquid dilu­ent to a mixing chamber of a device to form a first mixture;opening first valve means selectively control­ling passage of said first mixture from said mixing chamber to a hydrostatically connected mixture isolat­ing chamber, wherein a hydrostatically determined por­tion of said first mixture enters said mixture isolat­ing chamber;closing said first valve means, thereby iso­lating said portion from the remainder of said mixture;transferring said portion to a mixing chamber for dilution with a predetermined volume of a second diluent to form a second mixture.
