A device for the rapid qualitative or quantitative assay of entities having a biological activity.

Abstract

The invention relates to a novel assay for the determination of entities having biological activity and to a device for carrying out such determinations. The assay is a very sensitive one and quantities in the picogram per millimeter range can be determined. The assay is based on chrono-coulometric measurements with sequential measurements of a plurality of samples being carried out with the aid of a multiplexer, in combination with a potentiostat, there being provided suitable electrodes and means for applying a predetermined voltage. One of the electrodes is advantageously a glassy carbon electrode, carbon felt or cloth or carbon paper. Amongst biologically active entities there are antibodies/antigens; hormones/receptors ; nuclotides/ nucleotide probes, one of the members of such a pair being immobilized on an electrode surface, being tagged with an enzyme providing a signal in coulometric measurements.

    A device for the rapid qualitative or quantitative assay of entities having a biological activity by sequential determination of a plurality of samples by means of a plurality of enzyme electrodes immersed in a common solution in a vessel, there being provided means for effecting chrono-coulometric sequential measurements by employing a multiplexer, a potentio­stat and microcomputer, there being provided in said vessel a reference electrode and a counterelectrode, means being provided for the application of a predetermined voltage during each such measurement, the measurement being that of the current passing during the actual measurement and its evaluation to indicate the quantity of the measured moiety.

    A device according to any of claims 1 to 3, where the active species is bonded to the carbon felt, paper or cloth by carbodiimide.

    A device according to any of claims 1 to 3, where there are provided 6 or more electrodes with suitable multiplexing means.

    An assay for the qualitative or quantitative deter­mination of biologically active molecules are selected from antibodies/antigens;hormones/receptors;nuclotides/nucleo­tide probes, one of such pairs being immobilized on the electrode surface of one of the electrodes of a device claimed in any of claims 1 to 6, said entity being tagged with an enzyme adapted to provide a suitable signal in a coulometric measurement.
