A method for the preparation of liposomes, a pack for use therein and a composition for use in preparing a liposomal material.

Abstract

The invention provides a method for the preparation of liposomes by preparing a solution of a lipid component in an organic solvent and initially combining the thus-formed solution with a second component comprising water to form a reservoir of combined material which affords a multiple-dose system. Then a portion of an intimate mixture formed from the combination can be passed under pressure through an orifice to produce an aerosol spray containing liposomes. Generally an active material which is biologically-active and/or useful in the treatment or care of the human or animal body or of plants may be included in the combination. Also provided by the invention is a pack comprising at least a first chamber containing said combination. The first chamber and/or a second chamber includes at least one propellant material, and the pack includes an arrangement for dispensing the combination through an orifice. In a modification the pack comprises at least a first chamber containing a solution of a lipid component in an organic solvent and means to supply an aqueous component to the chamber. In addition, the invention provides a composition for use . in preparing a plurality of unit doses comprising liposomes based on a mixture of said lipid and aqueous components.

    A method for the preparation of liposomes, which method comprises preparing a solution of a lipid component in an organic solvent combining the thus-formed solution with a second component comprising water, forming an intimate mixture from the combination, and passing a portion of the combination as said mixture under pressure through an orifice, thereby to produce an aerosol spray containing liposomes.

    A method according to any one of the preceding claims, wherein the lipid component comprises a phospholipid material and is chosen so as to provide a positively charged, a negatively charged or a neutral liposome, and preferably the phospholipid comprises a phosphatidyl choline, a phosphatidyl ethanolamine or a phosphatidyl serine, and the lipid component comprises either one or more of those materials alone or one or more of those materials together with cholesterol and/or at least one of dicetyl phosphate, phosphatidic acid or stearylamine.

    A method according to any one of the preceding claims, wherein the aqueous component includes a buffering material.

    A method according to any one of the preceding claims, wherein the spraying pressure is produced by at least one aerosol propellant

    A method according to any one of claims 2 to 7, wherein the mixture is sprayed in a manner which provides a metered dose of the said active material.

    A pack for use in preparing a liposomal aerosol, which pack comprises at least a first chamber containing a solution of a lipid component in an organic solvent and an aqueous component, the first chamber and/or a second chamber including at least one propellant material, and the pack including an arrangement for dispensing through an orifice a mixture comprising the lipid solution and aqueous component as a spray under pressure developed by the propellant material or materials.

    A pack according to any one of claims 9 to 11, wherein the lipid solution, aqueous component, propellant(s), and any said active material are as defined in any one of claims 3 to 7.

    A composition according to any one of claims 13 to 15, wherein the lipid solution, aqueous component, propellant-(s), and any said active material are as defined in any one of claims 3 to 7.

A modification of a pack for use in preparing a liposomal aerosol according to any one of claims 9 to 12, which pack comprises at least a first chamber containing a solution of a lipid component in an organic solvent, means to supply an aqueous component to the chamber, the first chamber, a second chamber and/or the aqueous component including at least one propellant material, and the pack including an arrangement for dispensing through an orifice a mixture of said solution and aqueous component as a spray under pressure developed by the propellant material or materials.
