Abstract
Glycerol derivatives of the general formula: (wherein R1 represents an alkyl group of 6 to 22 carbon atoms or an alkyl group of 2 to 5 carbon atoms substituted by a phenyl group, A represents a single bond or an oxygene atom, R2 represents an alkyl group of 1 to 20 carbon atoms, an alkenyl group of 2 to 20 carbon atoms when A represents a single bond or an alkenyl group of 3 to 20 carbon atoms when A represents an oxygene atom, B represents an oxygene atom or a carbonyloxy group (i.e. -O-CO- group; with the proviso that in which a carbonyl group is attached to R3 and an oxa-group is attached to a carbon atom at the third position of the glycerol skeleton), R3 represents an alkylene group of 1 to 12 carbon atoms and R4 represents a group of the formula : (in which R5 represents an alkyl group of 1 to 6 carbon atoms or a phenyl group, R6 represents an alkyl group of 1 to 6 carbon atoms and represents an anion of an acid; with the proviso that when plural R6 are attached to a nitrogen atom, they may be same or different to each other)), and acid addition salt thereof, possess antagonistic effect on PAF, hypotensive effect like PAF, inhibitory effect on phospholipase and inhibitory effect on growth of tumour cell or induction of differentiation, and are, therefore, useful as a platelet aggregation inhibitor, anti-asthmatic agent, anti-allergic agent, anti-inflammatory agent, hypotensive agent and anti-tumour agent for mammals.

0001The present invention is concerned to new glycerol derivatives. More particularly, this invention is concerned to new glycerol derivatives having antagonistic effect on platelet activating factor (abbreviated as PAF hereafter), hypotensive effect like PAF, inhibitory effect on phospholipase and further, inhibitory effect on growth of tumour cell or induction of differentiation, and, furthermore, this invention is concerned to the process for their preparation and pharmaceutical composition comprising them as active ingredients.
0002Recently, the study on the release reaction of active amine dependent on leukocyte from platelet, has been energetically carried out. As a result, a substance strongly inducing platelet aggregation was discovered and it was named as PAF (cf. J. Immunol., 106, 1244 (1971), J. Exp. Med., 136, 1356 (1972) and Nature, 249, 581 (1974)) . Thereafter, it was confirmed that PAF exists in various animals including human beings. In 1979, its chemical structure was identified (cf. J. Biol. Chem., 254, 9355 (1979) and C.R. Acad. Sci., Serie D (Paris), 289, 1017 (1979)) and has turned out to be a mixture of two alkylphospholipids of the following general formula.wherein n is 15 and 17 (cf. J. Biol. Chem., 255, 5514 (1980)) : <chemistry id="chem0001" num="0001"><img file="EP0142333A2_D0001.tif" /></chemistry>(wherein n represents 15 or 17).
0003It was found that PAF has not only strong secretory effect on platelet aggregation known at that time, but also powerful hypotensive effect and bronchocontracting effect. PAF is considered to be one of platelet aggregating factors in human beings, and further, one of substances inducing allergy and inflammation. Accordingly, there is a great possibility that compounds having antagonistic effect on PAF (inhibitory effect on PAF) may become a platelet aggregation inhibitor, anti-asthmatic agent, anti-allergic agent or anti-inflammatory agent. Furthermore, PAF is considered to become a hypotensive agent without inhibitory effect on platelet 'aggregation by selecting hypotensive effect out of various effect.
0004Being interested in physiological effect on PAF, we have carried out widespread investigations in order to discover compounds similar to PAF in their chemical structure and possessing antagonistic effect on PAF or hypotensive effect like PAF. We have found that the above purpose can be accomplished by glycerol derivatives which are replaced the hydroxy group at the first position of the glycerol skeleton by an alkoxy group and are replaced the hydroxy group at the third position thereof by an alkoxy group substituted by a primary, secondary, tertiary or quaternary amine, preferably tertiary or quaternary amine.
0005For example, our European Patent Publication No. 94586 discloses the compounds of the general formula: <chemistry id="chem0002" num="0002"><img file="EP0142333A2_D0002.tif" /></chemistry>(wherein A<sup>1</sup> is -0- or -S-, B<sup>1</sup> is -0- or -S-, D<sup>1</sup> is -0- or -OCO-, <sub>R</sub><sup>11</sup> is alkyl, <sub>R</sub><sup>21</sup> is H, benzyl, acyl, N-substituted amido when <sub>B</sub><sup>1</sup> is -0-, or R<sup>21</sup> is alkyl, benzyl or alkoxy (thiocarbonyl) when <sub>B</sub><sup>1</sup> is -<sub>S</sub>-, <sub>R</sub><sup>31</sup> is alkylene and <sub>R</sub><sup>41</sup> is amino, mono- or dialkyl-substituted amino or trialkyl-substituted ammonium) and our European Patent Publication No. 109255 disclose the compounds of the general formula: <chemistry id="chem0003" num="0003"><img file="EP0142333A2_D0003.tif" /></chemistry>(wherein <sub>A</sub><sup>2</sup> is -<sub>0</sub>- or -OC<sub>O</sub>-, R<sup>12</sup> is alkyl, <sub>R</sub><sup>22</sup> is azido, amino, acylamino, alkoxycarbonylamino or ureido, R<sup>32</sup> is alkylene and R<sup>42</sup> is amino, mono- or dialkyl-substituted amino or trialkyl-substituted ammonium).
0006Now, we have found that by replacing functioned groups at the second position of the glycerol skeleton, hereinbefore described by an alkyl(oxy) or alkenyl(oxy) group and by limiting optionally substituted amino group attached to the end of the third position thereof to tertiary or quaternary amine, or amide, new glycerol compounds are more active in antagonistic effect on PAF than glycerol compounds previously proposed, having completed this invention.
0007On the other hand, glycerol compounds similar to those of the present invention in chemical structure are disclosed in the German Patent Publication No. 2835369. Broad scope of compound is disclosed therein and the compounds of the general formula: <chemistry id="chem0004" num="0004"><img file="EP0142333A2_D0004.tif" /></chemistry><sub>(</sub>wherein <sub>R13</sub> and <sub>R23</sub> each represents a normal alkyl or alkenyl group of 12 to 20 carbon atoms and Y and Z each represents an alkylene group of 2 to 4 carbon atoms or ..., R<sub>33</sub> represents a hydrogen atom, an alkyl group of 2 to 4 carbon atoms or ..., m, n and p each represents zero or 1 and the total number of m, n and p is zero or 1) are most related to our invention. (the definition not related to our invention is removed from the above description.)
0008As understood from the above definition, -NP.R<sub>3</sub> in formula (A) in DE-2835369 represents only a primary or secondary amine, which <sub>R</sub><sup>4</sup> in the general formula (I) hereinafter described, of the present invention represents a tertiary or quaternary amine.
0009Furthermore, the distinguished difference between the present invention and the invention disclosed in DE-2835369 is utility of glycerol compounds. That is, compounds of the present invention have antagonistic effect on PAF and hypotensive effect like PAF and, therefore, are used as platelet aggregation inhibitor, anti-asthmatic agent, anti-allergic agent and anti-inflammatory agent, whereas compounds in DE-2835369 have stimulating activity of production of interferon and are useful for prophylaxis of viral disease.
0010As discussed above, the present invention is remarkably different from the invention disclosed in DE-2835369 in chemical structure and their utility. The present invention is not anticipated and not obvious from DE-2835369.
0011Furthermore, it has been found that the compounds of the present invention of the general formula (I) have inhibitory effect on phospholipase A<sub>2 </sub>and phospholipase C. Phospholipase A<sub>2</sub> and phospholipase C dependent on calcium in a living body are concerned in the mechanism for the release of arachidonic acid from phospholipids. It is known that arachidonic acid is released by physiological stimulus and then becomes incorporated into the metabolic routes of prostaglandins and is finally metabolized to thromboxane A<sub>2 </sub>having a strong effect on platelet aggregation, to other prostaglandins or to various leukotrienes being a significant bronchostrictor in an asthmatic paroxysm. Accordingly, it is considered that compounds having an inhibitory effect on phospholipase A<sub>2</sub> and phospholipase C, which are concerned in the mechanism for the release of arachidonic acid, can become a platelet aggregation inhibitor, anti-asthmatic agent, anti-allergic agent or anti-inflammatory agent, based on the inhibition of phospholipase A<sub>2</sub> and phospholipase C, having a different mechanism from PAF antagonist and further, being different from conventional non-steroid anti-inflammatory agents represented by aspirin based on the inhibition of cyclooxygenase.
0012Furthermore, the compounds of the present invention of the general formula (I) show inhibitory effect on growth of tumour cell and induction of differentiation and, therefore, are may be useful as anti-tumour agents.

0015Examples of an alkyl group of 6 to 22 carbon atoms represented by R in the general formula (I) are hexyl, heptyl, octyl, nonyl, decyl, undecyl, dodecyl, tridecyl, tetradecyl, pentadecyl, hexadecyl, heptadecyl, octadecyl, nonadecyl, eicosyl, heneicosyl and docosyl group and isomers thereof; an alkyl group of 14 to 18 carbon atoms is preferred and hexadecyl group is most preferred.
0016Examples of an alkyl group of 2 to 5 carbon atoms substituted by a phenyl group, represented by R are ethyl, propyl, butyl and pentyl group and isomers thereof substituted any one of hydrogen atoms thereof by a phenyl group.
0017In R<sup>2</sup> of the general formula (I), examples of the alkyl group of 1 to 20 carbon atoms are methyl, ethyl, propyl, butyl, pentyl, hexyl, heptyl, octyl, nonyl, decyl, undercyl, dodecyl, tridecyl, tetradecyl, pentadecyl, hexadecyl, heptadecyl, octadecyl, nonadecyl and eicosyl group and isomers thereof, examples of an alkenyl group of 2 to 20 carbon atoms when A represents a single bond, are the groups which have a double bond between two carbon atoms in the alkyl group enumerated above (except methyl group) and examples of an alkenyl group of 3 to 20 carbon atoms when A represents an oxygen atom, are the groups which have a double bond between two carbon atoms except the carbon atom attached to an oxygen atom represented by A in the alkyl group enumerated above (except methyl and ethyl group); the alkyl group of 1 to 6 carbon atoms is preferred and allyl group is most preferred in the alkenyl group.
0018Examples of the alkylene group of 1 to 12 carbon atoms represented by R<sup>3</sup> in the general formula (I) are methylene, ethylene, trimethylene, tetramethylene, pentamethylene, hexamethylene, heptamethylene, octamethylene, nonamethylene, decamethylene, undecamethylene and dodecamethylene group and isomers thereof; the alkylene group of 3 to 8 carbon atoms is preferred and heptamethylene group is most preferred.
0019In R <sup>4</sup> of the general formula (I), examples of the alkyl group of 1 to 6 carbon atoms represented by R<sup>5</sup> or R are methyl, ethyl, propyl, butyl, pentyl and hexyl group and isomers thereof; every group is preferred. The anion of an acid represented by <chemistry id="chem0007" num="0007"><img file="EP0142333A2_D0007.tif" /></chemistry>mean the anion of a pharmaceutically-acceptable inorganic or organic acid, and examples of them are the anion of an inorganic acid such as hydrochloric acid, hydrobromic acid, hydroiodic acid, sulfuric acid, phosphoric acid, nitric acid, or the anion of an organic acid such as acetic acid, lactic acid, tartaric acid, benzoic acid, citric acid, methanesulfonic acid, ethanesulfonic acid, benzenesulfonic acid, toluenensulfonic acid, isethionic acid; more prefereable anion is a halogen ion, i.e. chlorine, bromine, iodine ions, or methanesulfonic acid or p-toluenesulfonic acid.
0020Preferred R<sup>4</sup> is the group of the formula: -N(R<sup>6</sup>)- or <chemistry id="chem0008" num="0008"><img file="EP0142333A2_D0008.tif" /></chemistry> (in which all the symbols are as hereinbefore defined), and the most preferred R<sup>4</sup> is the group of the formula : <chemistry id="chem0009" num="0009"><img file="EP0142333A2_D0009.tif" /></chemistry>
0021The compounds of the general formula (I) of the present invention have at least one asymmetric center, i.e. the carbon atom at the second position of the glycerol skeleton. When the alkyl moiety of various substituents represented by <sub>R</sub><sup>1</sup>, R<sup>2</sup>, R and R<sup>4</sup> represents a branched-chain, there is a possibility that other asymmetric centers are occurred. However the compounds of the general formula (I) in the present invention are concerned with all isomers or mixtures thereof which result from asymmetric centers.
0022According to a feature of the present invention, compounds of the general formula (I), wherein B represents an oxygen atom and R represents a group of the formula: <chemistry id="chem0010" num="0010"><img file="EP0142333A2_D0010.tif" /></chemistry> (in which Y<sup>2</sup> represents a halogen atom, alkylsulfonyloxy group or arylsulfonyloxy group and the other symbols are as hereinbefore defined), i.e. compounds of the general formulae: <chemistry id="chem0011" num="0011"><img file="EP0142333A2_D0011.tif" /></chemistry>and <chemistry id="chem0012" num="0012"><img file="EP0142333A2_D0012.tif" /></chemistry>(wherein the various symbols are as hereinbefore defined) may be prepared by the reaction of a compound of the general formula: <chemistry id="chem0013" num="0013"><img file="EP0142333A2_D0013.tif" /></chemistry>(wherein R<sup>7</sup>. represents an alkylsulfornyl group, a methanesulfonyl group (i.e. a mesyl group) is preferred or a substituted or unsubstituted arylsulfonyl group, a p-toluenesulfonyl group (i.e. tosyl group) is preferred and the other symbols are as hereinbefore defined) or a compound of the general formula: <chemistry id="chem0014" num="0014"><img file="EP0142333A2_D0014.tif" /></chemistry>(wherein Y represents a halogen atom and the other symbols are as hereinbefore defined), respectively, with a compound of the general formula: <chemistry id="chem0015" num="0015"><img file="EP0142333A2_D0015.tif" /></chemistry>(wherein R<sup>6</sup> is as hereinbefore defined) in a suitable organic solvent, e.g. a lower alkanol such as methanol, ethanol or isopropylalcohol, or dimethylformamide or the mixture thereof, at a temperture from ambient to a reflux temperature of the solvent.
0023The compounds of the general formula (I), wherein B represents a carbonyloxy group and R<sup>4</sup> represents as a group of the formula : -N(R<sup>6</sup>)<sub>2</sub> (in which R<sup>6</sup> is as hereinbefore defined), i.e. compound of the general formula: <chemistry id="chem0016" num="0016"><img file="EP0142333A2_D0016.tif" /></chemistry>(wherein all the symbols are as hereinbefore defined) may be prepared by esterification of a compound of the general formula: <chemistry id="chem0017" num="0017"><img file="EP0142333A2_D0017.tif" /></chemistry>(wherein all the symbols are as hereinbefore defined) with a compound of the general formula: <chemistry id="chem0018" num="0018"><img file="EP0142333A2_D0018.tif" /></chemistry>(wherein all the symbols are as hereinbefore defined).
0024The esterification may be carried out in an inert organic solvent such as ethyl acetate, benzene, ethyl ether, chloroform, methylene chloride or tetrahydrofuran using dicyclohexylcarbodiimide, trifluoroacetic acid anhydride or N,N'-carbodiimidazole as a condensing agent at a temperature from 0°C to 50°C (preferably at ambient). When dicyclohexyl- carbodiimide is used as a condensing agent, 4-dimethylaminopyridine or pyridine etc. is used as a agent of promoting reaction.
0025The compound of the general formula (I), wherein B represents a carbonyloxy group and R <sup>4 </sup>represents a group of the formula : <chemistry id="chem0019" num="0019"><img file="EP0142333A2_D0019.tif" /></chemistry> (in which all the symbols are as hereinbefore defined), i.e. compound of the general formula: <chemistry id="chem0020" num="0020"><img file="EP0142333A2_D0020.tif" /></chemistry>(wherein all the symbols are as hereinbefore defined) may be prepared by the reaction of a compound of the general formula: <chemistry id="chem0021" num="0021"><img file="EP0142333A2_D0021.tif" /></chemistry>or <chemistry id="chem0022" num="0022"><img file="EP0142333A2_D0022.tif" /></chemistry>(wherein all the symbols are as hereinbefore defined) with a compound of the general formula : N(R<sup>6</sup>)<sup>3</sup> (wherein R<sup>6</sup> is as hereinbefore defined). The reaction may be carried out under the same condition as mentioned above for the reaction of a compound of the general formula (IIa) or (IIIa) with a compound of the general formula : N(R<sup>6</sup>)<sub>3</sub>.
0026The compound in which R4 represents a group of the formula -NHCOR<sup>5</sup> (in which R<sup>5</sup> is as hereinbefore defined), i.e. compound of the general formula : <chemistry id="chem0023" num="0023"><img file="EP0142333A2_D0023.tif" /></chemistry>(wherein all the symbols are as hereinbefore defined) may be prepared by acylation of a compound of the general formula (I) wherein R<sup>4</sup> represents an amino group, i.e. compound of the general formula: <chemistry id="chem0024" num="0024"><img file="EP0142333A2_D0024.tif" /></chemistry>(wherein all the symbols are as hereinbefore defined) by a method known per se. Suitable acylation may be carried out by using a corresponding acyl chloride, benzoyl chloride or acid anhydride in the presence of a tertiary amine such as pyridine or triethylamine in an inert organic solvent such as methylene chloride or in the absence of a solvent at a temperature below 50°C.
0027The compounds of the general formulae (IIa), (IIIa), (IIb), (IIIb) and (IV) may be prepared by the series of reactions depicted schematically below in Scheme I. In Scheme I, THP represents a tetrahydropyran-2-yl group, and the other symbols are as hereinbefore defined. <chemistry id="chem0025" num="0025"><img file="EP0142333A2_D0025.tif" /></chemistry>
0028In Scheme I, each step can be effected by using methods known per se. For example, step (a) and (d) are a reaction for eliminating THP group, and may be carried out by using a mixture of tetrahydrofuran, acetic acid and water, p-toluenesulfonic acid and anhydrous methanol or dilute hydrochloride and tetrahydrofuran at ambient or by heating.
0029Step (b) may be carried out by reacting a compound of the general formula (IV) wherein B represents an oxygen atom, with a compound of the general formula: <chemistry id="chem0026" num="0026"><img file="EP0142333A2_D0026.tif" /></chemistry>or <chemistry id="chem0027" num="0027"><img file="EP0142333A2_D0027.tif" /></chemistry>(wherein X represents a halogen atom and the other symbols are as hereinbefore defined) in an organic solvent such as dimethylformamide or tetrahydrofuran in the presence of a hydride of alkali metal such as sodium hydride at a temperature from ambient to 80°C.
0030When B represents a carbonyloxy group, it may be carried out by esterizing a compound of the general formula (IV) with a compound of the general formula: <chemistry id="chem0028" num="0028"><img file="EP0142333A2_D0028.tif" /></chemistry>(wherein all the symbols are as hereinbefore defind) as mentioned above.
0031Step (c) may be carried out by reacting a compound of the general formula (IV) wherein B represents an oxygen atom, with a compound of the general formula: <chemistry id="chem0029" num="0029"><img file="EP0142333A2_D0029.tif" /></chemistry>or <chemistry id="chem0030" num="0030"><img file="EP0142333A2_D0030.tif" /></chemistry>(wherein all the symbols are as hereinbefore defined) under the same condition as described in step (c).
0032When B represents a carbonyloxy group, it may be carried out by esterizing a compound of the general formula (IV) with a compound of the general formula: <chemistry id="chem0031" num="0031"><img file="EP0142333A2_D0031.tif" /></chemistry>(wherein all the symbols are as hereinbefore defined) as mentioned above.

These effect were confirmed by standard laboratory test and, for example, antagonistic effect on PAF was confirmed by the following screening system.
(1) Inhibitory effect on PAF-induced platelet aggregation
0053Whole blood of male guinea-pig was collected with a 3.8%. aqueous solution of trisodium citrate in a proportion of 9:1 (v/v) and the mixture was centrifuged for 10 minutes at room temperature (120 x g) to obtain platelet rich plasma. Using obtained plasma, PAF (10nM)-induced aggregation was determined by the absorbancy method using an aggregometer of Born (cf. J. Physiol., 162, 67 (1962)) .
0054The evaluation is conducted using the concentration of compounds of the present invention required to produce a 50% inhibition of each effect, i.e. IC<sub>50</sub>. The results are shown in the following table. <tables id="tabl0001" num="0001"><img file="EP0142333A2_D0055.tif" /></tables>
0055On the other hand, it was confirmed that the acute toxicity of all extent of the compounds of the present invention was more than 50 mg/kg by intravenous administration. Therefore, glycerol derivatives of the present invention may be considered to be sufficiently safe and suitable for medical use. For example, the mortality (dead/used) in glycerol derivatives of the present invention, i.e. compounds prepared in Example 2(b), 5, 5(a) and 5(b) was 2/5, 0/5, 0/5 and 0/5 respectively, when each glycerol compound was intravenously administered to tail vein in ten mice at the dose of 50 mg/kg.
0056For the purpose hereinbefore described, glycerol derivatives of the general formula (I), or non-toxic acid addition salts thereof may be normally administered systemically or partially, usually by oral or parenteral administration. The dose to be administered is determined depending upon age, body weight, symptom, the desired therapeutic effect, the route of administration, and the duration of the treatment etc. In the human adult, the doses per person are generally between 1 mg and 1 g, preferably between 20 mg and 200 mg by oral administration up to several times per day, and between 100 g and 100 mg, preferably between 1 mg and 10 mg by parenteral administration up to several times per day.
0057As mentioned above, the doses to be used depend on various conditions. Therefore, there are cases in which doses lower than the ranges specified above and doses greater than the ranges specified above, may be used.
0058Solid compositions according to the present invention for oral administration include compressed tablets, dispersible powders and granules. The compositions for oral administration also include capsules of absorbable material such as gelatin.

0065Under an atmosphere of argon, 3.5g of sodium metal and a solution of 5g of malonic acid diethyl ester on the market in 20ml of ethanol were added to 80ml of ethanol, and 4g of n-propyl bromide was added thereto and then the mixture was refluxed overnight. To the reaction mixture were added a dilute aqueous solution of hydrochloride and ice, and the mixture was extracted with ether. The extract was washed with a saturated aqueous solution of sodium bicarbonate and water, successively, dried and concentrated under reduced pressure. 0072To 10ml of methylene chloride, 390mg of 2-dehydroxy-2-n-propylglycerol (prepared in Reference Example 2), 252mg of 2,3-dihydropyran and 3mg of p-tolulenesulfonic acid were added and the mixture was stirred for an hour at ambient. The reaction mixture was neutralized by adding a saturated aqueous solution of sodium bicarbonate and the mixture was extracted with ethyl acetate. The extract was washed with water and a saturated aqueous solution of sodium chloride successively, dried and concentrated under reduced pressure. 0073Under an atmosphere of argon, to 10ml of N,N-dimethylformamide, 73mg of sodium hydride and 199mg of 2-n-propyl-3-0-(tetrahydropyran-2-yl)glycerol compound (prepared in Reference Example 3) were added. After the mixture was stirred for an hour at 60°C, cooled by ice water and 600mg of hexadecyl bromide was added thereto and the mixture was stirred for 10 minutes. After the mixture was warmed up to 70°C and stirred for 4 hours, 0.5ml of water was added thereto and the mixture was concentrated under reduced pressure. The residue was purified by column chromatography on silica gel (eluent ; n-hexane : ethyl acetate = 10 : 1) to give 510mg (impurities were included) of the title compound having the following physical characteristic: <ul id="ul0007" list-style="none"><li>TLC(n-hexane : ethyl acetate = 5 : 1) : Rf . 0.71.</li></ul>
