Abstract
The solution concerns a mouse lymphocyte hybridoma producing an antibody against human leukocyte acid-labile interferon alpha, deposited in the hybridoma collection of the Institute of Virology of the Slovak Academy of Sciences in Bratislava under the designation VÚ IFN AL6. The monoclonal antibody of the VÚ IFN AL6 hybridoma is suitable for determining the presence of acid-labile interferon alpha based on the inhibition of its biological activity in interferon-neutralization tests. The monoclonal antibody is also suitable for the preparation of sera specific to mouse immunoglobulin of the IgG2a subclass.

- 1PREDMET VY Myší lyinfocytárny hybridóm VÚ IFN AL6, produkujúci monoklonálnu protilátku podtrie- 2 génnu protilátku, tzv. monoklonálnu protilátku, ktorá je schopná specificky reagovať s příslušným interferónom. Hybridóm VÚ IFN AL6 možno kultivovat’ in vitro v médiách vhodných pre živočišné buňky, alebo in vivo v peritoneálnrj dutině myší kmeňa BALB/c. Z konzerv, uchovaných v kvapalnoin dusíku, možno zahájit’ produkciu protilátky bez ďalej imunizácic antigénom. Příklad Pre účel pomnoženia hybridómových buniek in vivo sa aplikovalo 5 χ 106 buniek do peritomeálnej dutiny myší. Pre lepšie uchytenie aplikovaných buniek bola myš 15 dní před prenosom buniek hybridómu premedikovaná parafínovým olejom (0,5 ml intraperitoneálne). Po 20 dňoch rastu hybridómu v peritoneálnej dutině bola myš zabitá a vyprodukovaná ascitická tekutina odobratá. Celkom sa získalo 7 ml ascitickej tekutiny, ktorá obsahovala 8 mg/ml imunoglobulínu. Ascitická tekutina, obsahujúca produkt hybridómu VÚ IFN AL6 Specificky inhibovala antivírusovú a antiproliferatívnu aktivitu 1’udského acidolabilného interferónu alfa. In vitro rastů hybridómy ako polosuspenzná kultúra. Majú tvar (gufatý) a vefkosť charakteristické pre myelómové buňky, obsahujú fúzované buňkové jadrá, sú aneuploidné. Buňky hybridómu VÚ IFN AL6 majú ultraštruktúrny obraz typických myelómových buniek, kde prevažujúcou organelou sú vofné a na membrány viazané polyribozómy. Základným kultivačným médiom je Dulbeccova modifikácia Eaglovho miniinálneho esenciálneho média (Dulbecco, R. a Freeman, C., Virology 8 /1959/, 396). Toto médium, označované ako DMEM, je pre kultiváciu hybridómu doplněné gentamycínom a inaktívovaným koňským sérotn (10%), Elán Cloně, Bratislava. Hybridóm je kultivovaný pri 37 °C — jeho středná generačná doba je přibližné 24 hod. Produkovaná protilátka je monoklonálny imunoglobulín podtriedy IgG2a. Hybridóm VÚ IFN AL6 móže byť priemyslovo využívaný ako zdroj protilátky proti 1’udskému leukocytárnemu acidolabilnému interferónu alfa pri kvantitatívnom alebo kvalitatívnom vyhodnocovaní přítomnosti acidolabilného interferónu alfa vo vyšetrovanom materiáli alebo ako zdroj protilátky jedinej podtriedy (lgG2a) pre přípravu sér Specifických pre uvedenú podtriedu, NÁLEZU dy IgG2a proti 1’udskému acidolabilnému interferónu alfa.

Additionally, antibodies to interferon are made by interferon injection repeatedly as an antigen in experimental animals, most commonly rabbits or sheep. The serum of such immunized animals, taken after a certain antigenic antigen time, serves as a source of antibodies used primarily for quantitative or qualitative analysis of the antigen - the interferon molecule - in research and immunodiagnostic practice. This procedure, called conventional immunization, has several disadvantages. In the sera of immunized animals there is a heterogeneous mixture of antibodies, the spectrum of which in each individual organism is unusual and unrepeatable. Generally, the organism will create, in addition to antibodies against the desired antigen, antibodies to impurities of the antigenic preparation that are required to remove from the serum by shedding. Thus, conventional batch serum production is difficult to standardize and is within a wide range of quality. For the production of each batch, a clean immunization antigen and other antigens should be prepared to remove ballast antibodies against impurities.
The aforementioned drawbacks of the above-mentioned and commonly used procedure are eliminated when a hybridoma producing a monoclonal antibody against the human acid-labile interferon alpha is deposited in the collection of hybridomas of the SIA Virology Institute in Bratislava, Mlýnská dolina 1, under the name of the IFN AL6.
