Synthesis of long-chain polyunsaturated fatty acids by recombinant cells

Abstract

The present invention relates to methods of synthesizing long-chain polyunsaturated fatty acids, especially eicosapentaenoic acid, docosapentaenoic acid and docosahexaneoic acid, in recombinant cells such as yeast or plant cells. Also provided are recombinant cells or plants which produce long-chain polyunsaturated fatty acids. Furthermore, the present invention relates to a group of new enzymes which possess desaturase or elongase activity that can be used in methods of synthesizing long-chain polyunsaturated fatty acids.

    CLAIMS:1. A Brassica plant cell for producing docosapentaenoic acid (DPA) and docosahexaenoic acid (DHA) in an esterified form as part of triacylglycerols in the plant cell, comprising a polynucleotide which encodes a Δ4 desaturase whose amino acid sequence is at least 95% identical to the sequence set forth as SEQ ID NO:4 or SEQ ID NO:33, operably linked to a promoter which directs expression of the polynucleotide in the plant cell;a polynucleotide which encodes a Δ5 elongase operably linked to a promoter which directs expression of the polynucleotide in the plant cell;and an exogenous desaturase which desaturates an acyl-CoA substrate.

    A process for producing DPA and DHA in an esterified form as part of triacylglycerols, the process comprising i) obtaining Brassica seed comprising a cell of claim 1 or claim 2, and ii) extracting triacylglycerols from the seed.

    A process for producing DPA and DHA in an esterified form as part of triacylglycerols, the process comprising i) obtaining Brassica seed comprising a cell according to any one of claims 5 to 7, and ii) extracting triacylglycerols from the seed.

    A substantially purified polypeptide selected from the group consisting of:i) a polypeptide comprising an amino acid sequence as provided in SEQ ID NO:4, and ii) a polypeptide comprising an amino acid sequence which is at least 70% identical to SEQ IDNO:4, wherein the polypeptide has Δ4 desaturase activity.

    An isolated polynucleotide comprising a sequence of nucleotides selected from the group consisting of:i) a sequence of nucleotides as provided in SEQ ID NO:12;and ii) a sequence of nucleotides encoding a polypeptide according to claim 10 or claim 11 ;wherein the polynucleotide encodes a polypeptide having Δ4 desaturase activity.

    A recombinant cell comprising an exogenous polynucleotide comprising a sequence of nucleotides selected from the group consisting of:i) a sequence of nucleotides as provided in SEQ ID NO: 12;and ii) a sequence encoding a polypeptide according to claim 10 or claim 11;wherein the polynucleotide encodes a polypeptide having Δ4 desaturase activity.

    A method of producing DHA in a cell or a plant, the method comprising i) culturing, under suitable conditions, a recombinant cell according to any one of claims 15 to 18, and/or ii) cultivating, under suitable conditions, a transgenic plant comprising at least one cell according to any one of claims 15 to 18, wherein the cell or plant produces DHA.

    A method of producing oil comprising DHA, comprising obtaining a transgenic plant or part thereof comprising at least one cell according to any one of claims 15 to 18, and extracting oil from said plant or part thereof, wherein the plant or plant part comprises DHA.

    A process for feeding fish comprising:a) obtaining a feedstuff which comprises oil extracted from transgenic Brassica seeds which comprise docosapentaenoic acid (DPA) and docosahexaenoic acid (DHA) in an esterified form as part of triacylglycerols in the seeds, wherein the Brassica seeds further comprise i) a polynucleotide which encodes a Δ4 desaturase whose amino acid sequence is at least 95% identical to the sequence set forth as SEQ ID NO:4 or SEQ ID NO:33, operably linked to a promoter which directs expression of the polynucleotide in Brassica seed cells;ii) a polynucleotide which encodes a Δ5 elongase operably linked to a promoter which directs expression of the polynucleotide in the Brassica seed cells;and iii) an exogenous desaturase which desaturates an acyl-CoA substrate;and b) administering the feedstuff to fish.

    A process for feeding fish comprising:a) obtaining a feedstuff which comprises oil extracted from transgenic Brassica seeds which comprise docosapentaenoic acid (DPA) and docosahexaenoic acid (DHA) in an esterified form as part of triacylglycerols in the seeds, wherein the seeds comprise an exogenous desaturase which desaturates an acyl-CoA substrate and a sequence of nucleotides selected from the group consisting of: i) a sequence of nucleotides as provided in SEQ ID NO: 12;and ii) a sequence of nucleotides encoding a polypeptide according to claim 10 or claim 11, wherein the sequence of nucleotides encodes a polypeptide with Δ4 desaturase activity;and b) administering the feedstuff to fish.
