Conjugate based systems for controlled drug delivery

Abstract

Conjugates which comprise a drug and a ligand which includes a first saccharide; wherein the conjugate is characterized in that, when the conjugate is administered to a mammal, at least one pharmacokinetic or pharmacodynamic property of the conjugate is sensitive to serum concentration of a second saccharide. Exemplary conjugates and sustained release formulations are provided in addition to methods of use and preparation.

    A conjugate comprising an insulin molecule conjugated to one or more ligands wherein at least one ligand comprises aminoethylfucose (AEF);wherein the conjugate is characterized in that, when the conjugate is administered to a mammal in the absence of an exogenous multivalent saccharide-binding molecule, at least one pharmacokinetic or pharmacodynamic property of the conjugate is sensitive to serum concentration of a saccharide in the serum of the mammal.

    The conjugate of any one of claims 6-19, wherein the two or more separate ligands are aminoethylfucose (AEF).

    A conjugate comprising an insulin molecule conjugated to aminoethylfucose (AEF).

    The conjugate of any one of claims 12-21, wherein the insulin molecule is conjugated via the Al amino acid residue.
