Abstract

Polypeptides and polynucleotides encoding same comprising at least one carboxy-terminal peptide (CTP) of chorionic gonadotrophin attached to a carboxy terminus of a coagulation factor. Pharmaceutical compositions comprising the polypeptides and polynucleotides of the invention and methods of using same are also disclosed.

    The polypeptide of any one of claims 1 or 3, wherein the amino acid sequence of said coagulation factor is as set forth in amino acids 39-444 of SEQ ID NO:9.

    The polypeptide of any one of claims 2 or 5, wherein the amino acid sequence of said coagulation factor is as set forth in amino acids 47-461 of SEQ ID NO:17.

    The polypeptide of any one of claims 1-6, wherein the sequence of at least one CTP comprises an amino acid sequence selected from the group consisting of:SEQ ID NO: 1 and SEQ ID NO: 2.

    The polypeptide of any one of claims 1-7, wherein at least one CTP comprises at least one glycosylation site.

    The polypeptide of any one of claims 1-8 wherein at least one CTP is attached to said coagulation factor via a linker.

    A method of extending the biological half life of a coagulation factor, comprising the step of attaching between one and three chorionic gonadotropin carboxy terminal peptides (CTPs) to the carboxy terminus of said coagulation factor, wherein no CTP is attached to the amino terminus of said coagulation factor, wherein said CTPs comprise SEQ ID NO:3, wherein said coagulation factor is Factor IX, thereby improving the biological half life of said coagulation factor.

    A method of improving the area under the curve (AUC) of a coagulation factor, comprising the step of attaching between one and five chorionic gonadotropin carboxy terminal peptides (CTPs) to the carboxy terminus of said coagulation factor, wherein no CTP is attached to the amino terminus of said coagulation factor, wherein said CTPs comprise SEQ ID NO:3, wherein said coagulation factor is Factor VII or Factor Vila, thereby improving the area under the curve (AUC) of said coagulation factor.

    A method of improving the area under the curve (AUC) of a coagulation factor, comprising the step of attaching between one and three chorionic gonadotropin carboxy terminal peptides (CTPs) to the carboxy terminus of said coagulation factor, wherein no CTP is attached to the amino terminus of said coagulation factor, wherein said CTPs comprise SEQ ID NO:3, wherein said coagulation factor is Factor IX, thereby improving the area under the curve (AUC) of said coagulation factor.

    A method of reducing the dosing frequency of a coagulation factor, comprising the step of attaching between one and five chorionic gonadotropin carboxy terminal peptides (CTPs) to the carboxy terminus of said coagulation factor, wherein no CTP is attached to the amino terminus of said coagulation factor, wherein said CTPs comprise SEQ ID NO:3, wherein said coagulation factor is Factor VII, or Factor Vila, thereby reducing said dosing frequency of said coagulation factor.

    A method of reducing the dosing frequency of a coagulation factor, comprising the step of attaching between one and three chorionic gonadotropin carboxy terminal peptides CA 02767503 2017-01-13 (CTPs) to the carboxy terminus of said coagulation factor, wherein no CTP is attached to the amino terminus of said coagulation factor, wherein said CTPs comprise SEQ ID NO:3, wherein said coagulation factor is Factor IX, thereby reducing said dosing frequency of said coagulation factor.

    The method of any one of claims 27, 29, or 31, wherein said Factor IX is an activated Factor IX.

    The method of any one of claims 26-32, wherein the sequence of at least one chorionic gonadotrophin carboxy terminal peptide comprises an amino acid sequence selected from the group consisting of:SEQ ID NO: 1 and SEQ ID NO: 2.
