Abstract

Small-volume oral transmucosal dosage forms or NanoTabs~ comprising a predetermined amount of a pharmaceutically active drug are provided. Exemplary applications include use of the NanoTabs~ to administer a drug for the treatment of acute, post-operative or breakthrough pain.

    CA 02636088 2016-04-14 WHAT IS CLAIMED IS:1. A unit dosage form for oral transmucosal administration to a subject, comprising: from about 2 to about 200 micrograms (meg) of sufentanil and a bioadhesive material, wherein said bioadhesive material provides for adherence to the oral mucosa of said subject, wherein said unit dosage form has a volume of less than 30 μΐ or a mass of less than 30 mg.

    The unit dosage form according to Claim 1, wherein said oral transmucosal administration is sublingual administration.

    The unit dosage form according to Claim 1, wherein said oral transmucosal administration is buccal administration.

    The unit dosage form according to Claim 1, wherein said unit dosage form comprises a dose of sufentanil selected from the group consisting of 5 meg, 10 meg, 15 meg, 20mcg, 30mcg, 40mcg, 50mcg, 60mcg, 70mcg and 80 meg.

    The unit dosage form according to Claim 1, wherein said unit dosage form has an erosion time from 30 seconds up to a time selected from the group consisting of 5 minutes, 10 minutes, 15 minutes and 30 minutes.

    The unit dosage form according to Claim 1, wherein a single oral transmucosal administration of said unit dosage form to the subject results in a bioavailability of greater than 65%.

    The unit dosage form according to Claim 1, wherein a single oral transmucosal administration of said unit dosage form to the subject results in a bioavailability of greater than 75%.

    The unit dosage form according to Claim 1, wherein a single oral transmucosal administration of said unit dosage form to the subject results in a bioavailability of greater than 85%. CA 02636088 2016-04-14

    The unit dosage form according to Claim 1, wherein a single oral transmucosal administration of said unit dosage form to the subject results in a bioavailability with a coefficient of variation of less than 30%.

    The unit dosage form according to Claim 1, wherein repeated oral transmucosal administration of said unit dosage form results in a bioavailability that is greater than the bioavailability following a single oral transmucosal administration.

    The unit dosage form according to Claim 1, wherein repeated oral transmucosal administration of said unit dosage form results in a bioavailability of greater than 65%.

    The unit dosage form according to Claim 1, wherein repeated oral transmucosal administration of said unit dosage form results in a bioavailability of greater than 75%.

    The unit dosage form according to Claim 1, wherein repeated oral transmucosal administration of said unit dosage form results in a bioavailability of greater than 85%.

    The unit dosage form according to Claim 1, wherein a single oral transmucosal administration of said unit dosage form results in a T max with a coefficient of variation of less than 40%.

    The unit dosage form according to Claim 1, wherein repeated oral transmucosal administration of said unit dosage form to the subject, results in a T max that is lower than the T max following a single oral transmucosal administration to said subject.

    The unit dosage form according to Claim 1, wherein an amount of drug selected from the group consisting of at least 55%, at least 60%, at least 65%, at least 70%, at least 75%, at least 80%, at least 85%, at least 90%, at least 95%, at least 98% and at least 99% of the total amount of drug in said unit dosage form is absorbed via the oral transmucosal route.     The unit dosage form according to Claim 1, wherein at least 60% of the total amount of drug in said unit dosage form is absorbed via the oral transmucosal route.
