Process for the preparation of activated polyethylene glycols

Abstract

A process for preparing activated polyethylene glycols is disclosed. In some embodiments, the process includes reacting a molten polyethylene glycol with an activator. In other embodiments, the process includes reacting a polyethylene glycol with an activator in the absence of a solvent. The process may be carried out in an inert gas atmosphere, at a temperature at least 10 Degree Celcius above the melting point of polyethylene glycol, and/or with the activator provided in molar excess of the polyethylene glycol. The invention further provides activated polyethylene glycols produced by this process and their use in a variety of pharmaceutical, medical, cosmetic and chemical applications.

A method for preparing an activated polyethylene glycol having the formula:Y-Q- (OCH2CH2)n-O-Q-Y wherein Q is selected from the group consisting of -C(O), -SO2-, >P(O), and -A-R-B- ;wherein A and B independently are -C (O), -SO2-, or >P(O) ;and R is selected from the group consisting of a lower alkyl group, a lower branched alkyl group, an aryl group, an aralkyl group, -(CHR])t-, -O-(CHRi)tO-, -S-(CHR])rS-, -O-(CHRi)t-S-, -S-(CHR])t-O-, -O-(CHRI)t-R2C=CR3-(CHR4)t-O-, -S-(CHRi)rR2C=CR3-(CHR4)rS-, -O-(CHRi)t-R2C=CR3-(CHR4) t-S-, and-S-(CHRt)n-R2C=CR3-(CHR4)t-O-;wherein Ri, R2, R3j and R> are independently selected from the group consisting of a hydrogen atom, a lower alkyl group, a lower branched alkyl group, an aryl group, and an aralkyl group;and t is 1, 2, or 3 ;Y is a leaving group;and n is an integer greater than 2;the method comprising the steps of : (i) providing a molten polyethylene glycol having the formula: M- (OCH2CH2)n-O-M wherein M is selected from the group consisting of H, Li, Na, K, Rb, and Cs;and n is as defined above;and (ii) reacting in the absence of a solvent the molten polyethylene glycol with an activator having the formula: Y-Q-X -33CA 02576152 2011-11-15 wherein X is a leaving group;Q and Y are as defined above.

    The method of any one of claims 1-3, wherein the activator is O2NPhOC(O)Cl.

    The method of any one of claims 1-4, wherein M is H.

    The method of any one of claims 1-5, wherein the reacting step is carried out at a temperature at least about 10°C above the melting point of polyethylene glycol.

    The method of any preceding claim wherein M is Η, n is an integer between 150 and 250, and the reacting step is carried out at a temperature in the range of about 6090°C.

    The method of any one of claims 1-7, wherein the reacting step is carried out in an inert atmosphere.

    The method of any one of claims 1-8, wherein the molar amount of the activator is in excess of the molar amount of the polyethylene glycol.
