Method for lyophilizing an active agent

Abstract

A process for lyophilizing a solution (50) of an active agent in a container (19) is provided. Solution of active agent is deposited into a container, the container is covered with a covering plate (60) and placed inside a lyophilizing apparatus. Lyophilization can be conducted to dryness by radiation, convection or both. Also provided is a device made by this process, an active agent-plastic administration device (e.g., a syringe (20)), containing an active agent in the form of lyophilized cake, and an array of such administration devices.

A method for lyophilizing a solution, the method comprising: depositing the solution into one or more first syringes;suspending a rack within a tub, the rack adapted to suspend the one or more first syringes in the tub;positioning a covering plate comprising one or more protuberances projecting from a surface to align with an opening of the one or more first syringes, the one or more protuberances adapted to fit inside at least a portion of the one or more first syringes;covering the one or more first syringes with the covering plate before placing the one or more first syringes and the covering plate inside a lyophilizing apparatus or covering the one or more first syringes with the covering plate after placing the one or more first syringes and the covering plate inside the lyophilizing apparatus, the one or more projecting protuberances engaged inside the at least a portion of the one or more first syringes;and at least partially lyophilizing the solution by cooling the solution and applying a vacuum to the solution, wherein at least partially lyophilizing the solution allows vapor to escape through an annular gap between each of the one or more protuberances and a side wall of each of the one or more first syringes, sufficient to retain a lyophilized medication in the one or more first syringes;sealing the one or more first syringes;contacting one or more second syringes with a diluent;contacting a matable luer-lock fitting of the one or more first syringes with the opposing matable luer-lock fitting of the one or more second syringes, sufficient to disengageably interlock the first and second syringes;fluidly mixing the lyophilized medication and diluent by alternating fluid tight movement of a first plunger positioned in the one or more first syringes and a second plunger positioned in the one or more second syringes.

    The method of any one of claims 1-3, wherein lyophilizing the solution produces about 5.0 mg to about 25.0 mg of lyophilizate in each of the one or more first syringes.

    The method of any one of claims 1-4, wherein lyophilizing the solution produces about 11.4 mg of lyophilizate in each of the one or more first syringes.

    The method of any one of claims 1-5, further comprising preparing the solution containing about 20 mg/ml to about 80 mg/ml of leuprolide acetate.

    The method of any one of claims 1-6, further comprising preparing the solution containing about 38 mg/ml of leuprolide acetate.

    The method of any one of claims 1-7, wherein depositing the solution into the one or more first syringes comprises depositing the solution containing about 0.1 ml to about 1.0 ml of leuprolide acetate into each of the one or more first syringes.

    The method of any one of claims 1-8, wherein depositing the solution into the one or more first syringes comprises depositing the solution containing about 0.3 ml of leuprolide acetate into each of the one or more first syringes.

    The method of any one of claims 1-9, wherein cooling the solution comprises cooling the solution by convection.

    The method of any one of claims 1-12, wherein cooling the solution comprises cooling the solution to a temperature between about 0°C and about -50°C.

    The method of any one of claims 1-13, wherein covering the one or more first syringes with the covering plate forms a lid preventing the escape of lyophilizate from the one or more first syringes during the lyophilization process.

    The method of any one of claims 1-14, wherein covering the one or more first syringes with the covering plate prevents contamination of lyophilizate from one of the one or more first syringes by the lyophilizate from another of the one or more first syringes when two or more of the first syringes are lyophilized simultaneously.

    The method of any one of claims 1-15, further comprising removing the covering plate from the one or more first syringes after the lyophilization of the solution.

    The method of any one of claims 1-13, further comprising:removing the covering plate from the one or more first syringes after the lyophilization of the solution;and examining a surface of the covering plate for retained lyophilizate and discarding the one or more first syringes from which the lyophilizate came.

    The method of any one of claims 1-17, further comprising sealing the one or more first syringes containing the at least partially lyophilized pharmaceutical.

    The method of any one of claims 1-17, wherein sealing the one or more first syringes comprises inserting a plunger into each opening of said one or more syringes.
