Preparation of submicron sized nanoparticles via dispersion and solvent or liquid phase removal

Abstract

The present invention relates to a process for preparing submicron sized nanoparticles of a poorly water soluble compound by sonicating to evaporate a portion of the organic phase or by lyophilizing a dispersion or microdispersion of a multiphase system having an organic phase and an aqueous phase, the organic phase havingthe poorly water soluble organic compound therein. The method is preferably used to prepare nanoparticles of a poorly water soluble, pharmaceutically active compound suitable for in vivo delivery, particularly by parenteral routes.

A process for preparing submicron sized particles comprising the steps of: providing a crude dispersion of a multiphase system having an organic phase and an aqueous phase, the organic phase having a pharmaceutically active compound therein;adding at least first and second surface active compounds to the multiphase system;providing energy to the crude dispersion to form a fine dispersion;and freezing the fine dispersion to obtain a frozen dispersed system of particles in a solid state, the particles having a mean particle size of less than 500 nm, wherein the first surface active compound is selected from the group consisting of anionic surfactants, cationic surfactants, and nonionic surfactants, and the second surface active compound comprises a surface active biological molecule.
