Local delivery of fibrinolysis enhancing agents

Abstract

A method of preventing adhesions by topical administration of fibrinolysis e nhancing agents is described. The method uses a topicall y applied polymeric matrix for delivery of a fibrinolytic agent, preferably ur okinase, tPA, hirudin, or most preferably, ancrod. In th e most preferred embodiment, the matrix is extremely thin and is polymerized in sut i to form a biodegradable polymeric matrix. The matrix provides controlled release of the agent over a period of time effective to prevent surgical adhesions and is biodegradable, usually within the same time frame. Examples demonstrate that the combination of the matrix and the urokinase, tPA, birudin, or ancrod is effective in preventing surgical adhesions.

16 claims: 2 independent, 14 dependent

  1. 1
We claim:1. A composition for preventing tissue adhesions comprising an effective amount of a fibrinolysis enhancing agent in a topically applied biocompatible, biodegradable polymeric matrix, wherein the matrix is formed by gelling or solidifying a biocompatible, biodegradable polymer or ma'cromer on tissue, and wherein the matix is capable of conforming to the tissue and releasing the agent in a controlled manner over a period of time effective to prevent adhesions.
  2. 15
A composition for preventing tissue adhesions comprising a pharmaceutically acceptible carrier for topical administration to tissue and an effective amount of a fibrinolysis enhancing agent to inhibit adhesions, wherein the fibrinolysis enhancing agent is selected from the group consisting of urokinase, thrombolytic agents, proteases specifically cleaving fibrin, anti-plasmin inhibitors, and fibrin deposition blockers.
