Recombinant antibodies specific for tnf-.alpha.

Abstract

Recombinant, in particular humanised, e.g. humanised chimeric and CDR-grafted humanised, antibody molecules having specificity for human TNF.alpha., are provided for use in diagnosis and therapy. In particular the antibody molecules have antigen binding sites derived from murine monoclonal antibodies CB0006, CB0010,hTNF3 or 101.4. Preferred CDR-grafted humanised anti-hTNF.alpha. antibodies comprise variable region domains comprising human acceptor framework and donor antigen binding regions and wherein the frameworks comprise donor residues at specific positions. The antibody molecules may be used for therapeutic treatment of human patients suffering from or at risk of disorders associated with undesirably high levels of TNF, in particular for treatment of immunoregulatory and inflammatory disorders of septic, endotoxic or cardiovascular shock.

An antibody molecule which has specificity for human TNF.alpha. comprising a composite heavy chain and a complementary light chain, said composite heavy chain having a variable domain comprising predominantly human acceptor antibody heavy chain framework residues and donor antibody heavy chain antigen-binding residues, said donor antibody having specificity for said human TNF.alpha., wherein, according to the Kabat numbering system, in said composite heavy chain, amino acid residues 23, 24, 31 to 35, 49 to 65 and 95 to 100 at least are donor residues.

    The antibody molecule of claim 1, wherein amino acid residues 71, 73 and 78 in said composite heavy chain are additionally donor residues.

    The antibody molecule of claim 1 or claim 2, wherein at least one of amino acid residues 2, 4, 6, 25, 36, 37, 39, 47, 48, 93, 94, 103, 104, 106 and 107 are additionally donor residues.

    The antibody molecule of claim 1 or claim 2 wherein at least one of amino acid residues 9, 11, 41, 67, 69 (if 48 is different between donor and acceptor), 87, 88, 91, 108, 110 and 112 or both of at least one of the pairs of residues 1 and 3, 18 and 82 (if 67 is a donor residue), 20 and 80 (if 69 is a donor residue), 38 and 46 (if 48 is a donor residue) and 72 and 76 are additionally donor residues.

    The antibody molecule of claim 1 or claim 2, wherein amino acid residues 26 to 30 are additionally donor residues.

    The antibody molecule of claim 1 or claim 2, wherein said light chain is a composite light chain having a variable domain comprising predominantly human acceptor antibody light chain framework residues and donor antibody light chain antigen-binding residues, said donor antibody having 44 specificity for said human TNF.alpha., wherein, according to the Kabat numbering system, amino acid residues 24 to 34, 46, 47, 50 to 56 and 89 to 97 at least are donor residues.

    An antibody molecule which has specificity for human TNF.alpha. comprising a composite light chain and a complementary heavy chain, said composite light chain having a variable domain comprising predominantly human acceptor antibody light chain framework residues and donor antibody light chain antigen-binding residues, said donor antibody having specificity for said human TNF.alpha., wherein, according to the Kabat numbering system, in said composite light chain, amino acid residues 24 to 34, 46, 47, 50 to 56, and 89 to 97 at least are donor residues.
