Growth hormone antagonists

Abstract

The present invention relates to growth inhibitory antagonists of bovine growth hormone obtained by mutation of the thirdalpha helix of that protein. These novel hormones may be administered exogenously to animals, or transgenic animals may bemade that express the antagonist and thereby exhibited a reduced growth phenotype.

    CA 02102129 2002-06-17 - 47 The embodiments of the invention in which an exclusive property or privilege is claimed are defined as follows:1. Use of a growth hormone antagonist peptide or protein of at least 50 amino acids, comprising an alpha helix which is at least 50% but not entirely identical to the third alpha helix of bovine growth hormone (bGH) or human growth hormone (hGH), wherein at least one of the differences from said third alpha helix of bGH or hGH is at a residue corresponding to bGH 119 or hGH 120 , said peptide or protein being capable of antagonizing at least the biological activity of a human or animal growth hormone in the manufacture of a composition for the prevention or treatment of a condition of said human or animal, which is characterized by the level of said biological activity being excessive and wherein the condition is diabetes .

    Use of a recombinant DNA molecule capable of expressing in a suitable host cell, a growth hormone antagonist peptide or protein of at least 50 amino acids, comprising an alpha helix which is at least 50% but not entirely identical to the third alpha helix of bovine growth hormone (bGH) or human growth hormone (hGH), wherein at least one of the differences from said third alpha helix of bGH or hGH is at a residue corresponding to bGH 119 or hGH 120 , said peptide or protein being capable of antagonizing at least the biological activity of a human or animal growth hormone, in the manufacture of a composition for the prevention or treatment of a condition of said human or animal, which is characterized by the level of said biological activity being excessive and wherein the condition is diabetes.

    Use of a growth hormone antagonist peptide or protein of at least 50 amino acids, comprising an alpha helix which is at least 50% but not entirely identical to the third alpha helix of bovine growth hormone (bGH) or human growth hormone (hGH), wherein at least one of the differences from said third alpha helix of bGH or hGH is at a residue corresponding to bGH 119 or hGH 120 , said peptide or protein being capable of antagonizing at least the biological activity of a human or animal growth hormone in the manufacture of a composition for the prevention or treatment of a condition of said human or animal, which is characterized by the level of said biological activity being excessive and wherein the condition is excessive levels of serum cholesterol and the peptide or protein has a hypocholesterolemic effect.

    Use of a recombinant DNA molecule capable of expressing in a suitable host cell, a growth hormone antagonist peptide or protein of at least 50 amino acids, comprising an alpha helix which is at least 50% but not entirely identical to the third alpha helix of bovine growth hormone (bGH) or human growth hormone (hGH), wherein at least one of the differences from said third alpha helix of bGH or hGH is at a residue corresponding to bGH 119 or hGH 120 , said peptide or protein being capable of antagonizing at least the biological activity of a human or animal growth hormone, in the manufacture of a composition for the prevention or treatment of a condition of said human or animal, which is characterized by the level of said biological activity being excessive and wherein the condition is excessive levels of serum cholesterol and the peptide or protein has a hypocholesterolemic effect.

    Use of a growth hormone antagonist peptide or protein of at least 50 amino acids, comprising an alpha helix which is at least 50% but not entirely identical to the third alpha helix of bovine growth hormone (bGH) or human growth hormone (hGH), wherein at least one of the differences from said third alpha helix of bGH or hGH is at a residue corresponding to bGH 119 or hGH 120 , said peptide or protein being capable of antagonizing at least the biological CA 02102129 2002-06-17 - 49 activity of a human or animal growth hormone in the manufacture of a composition for the prevention or treatment of a condition of said human or animal, which is characterized by the level of said biological activity being excessive and wherein the biological activity is tumorigenic.

    Use of a recombinant DNA molecule capable of expressing in a suitable host cell, a growth hormone antagonist peptide or protein of at least 50 amino acids, comprising an alpha helix which is at least 50% but not entirely identical to the third alpha helix of bovine growth hormone (bGH) or human growth hormone (hGH), wherein at least one of the differences from said third alpha helix of bGH or hGH is at a residue corresponding to bGH 119 or hGH 120 , said peptide or protein being capable of antagonizing at least the biological activity of a human or animal growth hormone, in the manufacture of a composition for the prevention or treatment of a condition of said human or animal, which is characterized by the level of said biological activity being excessive and wherein the biological activity is tumorigenic.

    The use according to any one of claims 1 - 8, wherein said peptide or protein comprises amino acids corresponding to amino acids 96 - 133 of bGH or hGH.

    The use according to any one of claims 1-8, wherein said peptide or protein comprises alpha helices corresponding to helices 1 - 4 of bGH, hGH or porcine growth hormone (pGH).

    The use according to any one of claims 1-8, wherein said alpha helix is at least 80% identical to the third alpha helix of bGH or hGH.
