DNABII vaccines and antibodies with enhanced activity

Abstract

The disclosure provides methods and compositions that are useful to lessen and/or cure bacterial biofilms and treat diseases or disorders associated with biofilms using one or more novel polypeptide vaccines, antibodies, antibody fragments and compositions. Bacteria that cannot form functional biofilms are more readily cleared by the remainder of the host's immune system and/or traditional antibiotics.

A chimeric recombinant polypeptide comprising two or more polypeptides selected from: isolated conformational tip domains of a DNABII polypeptide, wherein the conformational tip domains consist of polypeptides selected from SEQ ID NOs: 13, 17 and 78-136, and an optional linker polypeptide linking the two or more polypeptides.

    An isolated host cell comprising one or more of:a. the recombinant polypeptide of claim 1 or 2;and/or b. the recombinant polynucleotide or the vector of claim 3.

    An antibody or an antigen binding fragment of an antibody that binds a recombinant polypeptide that comprises two conformational tip domains that consist of SEQ ID NO:13 and SEQ ID NO: 17, and optionally wherein the antibody or antigen binding fragment thereof further comprises a detectable and/or a purification label.

    A polynucleotide or a vector comprising a polynucleotide encoding the antibody or antigen binding fragment of any one of claims 5 to 7, and optionally further comprising a detectable and/or a purification label.

    An isolated host cell comprising one or more of:a. the polynucleotide or the vector of claim 8;and/or b. the antibody or antigen binding fragment of any one of claims 5 to 7.

    A method for preventing formation of, or disrupting a biofilm associated with an industrial process, comprising contacting the biofilm with an effective amount of one or more of an antibody or antigen binding fragment of any one of claims 5 to 7.
