Binary encoded sequence tags

Abstract

(57) The detector composite for using for the comprehensive analysis method of a summary nucleic acid sample and this method is indicated. This method is called a binary code-ized arrangement tag (BEST), and generates a series of nucleic acid fragmentation, It includes adding an adapter to the end containing the recognition site for cutting by the part of offset from a recognition site, generating the fragmentation which cuts the fragmentation and has two or more sticky ends, giving an index to the fragmentation, and dividing into a set based on the arrangement of a sticky end. Fragmentation is given to an index by adding an offset adapter to the newly generated end. Coupling of the various adapters is carried out to a respectively different sticky end. The end is limited, length (setting in a desirable example) is equal, and the obtained fragmentation which has the main arrangement of the origin from the nucleic acid molecule of a source of supply is a binary arrangement tag. This binary arrangement tag can be used by very many methods, and also can be analyzed. For example, this binary arrangement tag can be hybridized with a probe, and can be captured by carrying out a rye gate preferably by carrying out coupling. This probe -- an array -- or fixing on the bead which can be classified is desirable. The form of 1 of the BEST method called ornamentation auxiliary analysis (MAABST) of a binary arrangement tag evaluates ornamentation of the arrangement in a nucleic acid molecule by detecting differential cutting on the basis of the existence of ornamentation of a molecule.
