Continuous manufacturing procedure of microcapsules for extended release of water soluble peptides

A process for producing sustained-release microcapsules of water soluble peptides, usable in injectable preparations and with periods of adjustable release between 1 and 18 weeks in which emulsifies, continuously, an aqueous solution of an active peptide containing a substance retaining, in an oily solution of a biodegradable polymer dissolved in an organic solvent sparingly soluble in water, using for this purpose a first intensive agitation equipment, closed to ambient, fed by metering; this first water / oil emulsion, after being cooled, is fed by a metering to a second intensive agitation equipment closed to ambient, where it is emulsified continuously in a vehicle aqueous phase containing a hydrophilic protective colloid, which is fed by a continuous dosing; this second complex water emulsion / oil / water type continuously transported in a closed container where the solvent is evaporated by pressure reduction, producing the microcapsules consolidation; wherein said microcapsules, obtained by a W / O / W double and continuous emulsification, are subjected to the following operations on wet: sieved in suspension; centrifuged; They washed with water; and fractionated into suitable containers, dispersed in an aqueous medium containing a lyophilization excipient; then frozen in a freezer orbital shaking until temperatures less than -20 and lyophilized in the same containers.
