Probe composition and method
Abstract
(57) The method and structure for detecting 1 or the selected polynucleotide domain beyond it in summary target polynucleotide. In order to enforce this method, in the case, several different sequence probe pairs are added to target polynucleotide, Each probe pair of these elements is complementary to the contiguity portion of one selected target sequence in / including two polynucleotide probe elements / target polynucleotide in an order. In each probe pair, one of a probe element contains the non-polynucleotide polymer chain given to the probe pair which met the mobility of the characteristic electrophoresis in the sieving matrix, when connecting the element in an opposite. Other elements in this opposite contain a detectable reporter label. After a probe pair carries out hybrid formation by target polynucleotide, the polynucleotide which carried out hybrid formation is processed under an effective condition, in order to connect the end subunit of a target joint probe element, when the end subunit is an adjoining target base and base pair. The connected probe pair disconnects itself of target polynucleotide next, and is divided into a sieving matrix by an electrophoresis or chromatography.
