Efficient haploid cell sorting for flow cytometer systems

Abstract

A flow cytometric system (1) to classify haploid cells, specifically irradiable sperm cells, with an intermittent point radiation emitter (56). Embodiments include a beam manipulator (21) and separate parallel radiation beams directed at multiple nozzles (5). Differentiating sperm characteristics with increased resolution could allow differentiated sperm cells to be efficiently separated at high speeds and in subpopulations that have high purity. Claim 1: A method of sperm processing by flow cytometry comprising the steps of: stabilizing an envelope fluid; flowing said wrapping fluid into a nozzle; injecting irradiable sperm cells into said covering fluid; subjecting said irradiable sperm cells to multiple radiations for a first amount of time; terminating said multiple radiation from said irradiable sperm cells for a second amount of time; multiple exciting said irradiable sperm cells with said radiation; fluorescing from said excited sperm cells; detecting an amount of said fluorescence emitted from each of said sperm cells; evaluating said amount of fluorescence emitted from each of said sperm cells; selecting an electrical condition to be associated with each of said sperm cells in the flow of said envelope fluid; charging a stream of said irradiable sperm cells and said envelope fluid based on the inferred properties of each of said excited sperm cells in said envelope fluid flow; form a charged drop; isolating said charged drop from said covering fluid flow; diverting such charged drops; classify said sperm cells; and collecting said classified sperm cells.
